When evaluating cook time on a brisket, would I do so based on original weight or trimmed weight?

Trimmed weight. But remember, those "rule of thumb" times are very general in nature.

Trimmed.

The stuff you trimmed off isn't going into the cooker so it doesn't matter during the cook.
